题目 | The unique germinal compartments of human medial ganglionic eminence sustain interneuron production |
摘要 | Cerebral cortical inhibitory interneurons (INs) play an extremely important role in regulating and integrating neural network signals, and defects in the production of INs usually lead to neurodevelopmental abnormalities and the occurrence of a variety of psychiatric disorders. Although studies have shown that the molecular mechanism regulating INs differentiation and migration between species is conserved, this does not answer the reason why the diversity of cell types of human INs is significantly higher than that of mice, and the number and proportion of human cerebral cortex INs increased. We have observed the formation of nest-like structures (GCs: Germinal compartments) containing a large number of INs precursor cells in the hMGE, and their persistence throughout the long process of human embryonic development, suggesting that GCs may be the basic neurogenesis unit unique to human. However, due to the limitations of samples and technical means, the fine cell composition and physiological functions of GCs are still unclear. In this project, based on the previous research results, we will use spatial transcriptome and single-cell transcriptome sequencing and in situ micro-transplantation to conduct analysis and functional exploration of GCs, providing new possible mechanisms of human unique neurogenesis. |
